L'accademia era stata preceduta dalla Regia scuola di recitazione "Eleonora Duse", a sua volta nata nel 1921 da una costola dell'Accademia Nazionale di Santa Cecilia. Ne erano stati direttori Virginia Marini, Cesare Dondini e infine Franco Liberati e Silvio D'Amico vi insegnò "storia del teatro". Allievi della scuola erano stati Anna Magnani, Paolo Stoppa, Sergio Tofano, e Orazio Costa (diplomato nel 1932 e che in seguito frequentò anche i corsi di regia dell'accademia, diventandone insegnante dal 1945 per oltre vent'anni). Nel 1935 la scuola venne soppressa, contestualmente alla fondazione dell'accademia, con la direzione di Silvio D'Amico; insegnanti furono Tat'jana Pavlovna Pavlova per la regia, Luigi Almirante, Irma Gramatica e Gualtiero Tumiati per la recitazione, Virgilio Marchi per "scenotecnica e storia del costume", Teo Fasulo per danza, Valentino Amannato per scherma, Gino Viotti per trucco, Maria Labia per "impostazione al canto lirico" e lo stesso Silvio D'Amico per "storia del teatro". L'insegnamento della regia passò nel 1938 a Guido Salvini, che lo tenne fino al 1942. Furono ancora insegnanti di recitazione Nera Grossi Carini (fino al 1947), Mario Pelosini (fino al 1950) e Carlo Tamberlani (sostituito nel 1939 da Wanda Capodaglio), ai quali si aggiunse Alba Maria Setaccioli dal 1948). La danza fu in seguito affidata a Raja Garosci e in seguito ad Anna Berger, poi Avia de Luca e, un poco più a lungo, Rosa Mazzucchelli.
